Glucotoxicity and lipotoxicity induced beta-cell apoptosis in type 2 diabetes mellitus

چکیده
Type 2 Diabetes mellitus (T2DM) is a chronic disorder of impaired insulin secretion and insulin resistance leading to hyperglycemia. In addition to hyperglycemia, dyslipidemia is a characteristic feature of T2DM. Progressive deterioration of beta-cell mass and function are the core pathogenic mechanisms underlying diabetes. Persistent hyperglycemia and hyperlipidemia are the major factors for loss of pancreatic beta-cell mass through apoptosis. Glucose is a key physiological regulator of insulin secretion and small changes in glucose concentration exceeding the physiological range induces the pro-apoptotic signals including endoplasmic reticulum stress, mitochondrial dysfunction and oxidative stress. The role of noggin in regulation of high glucose-induced apoptosis and insulin secretion in INS-1 rat beta cells
Objective(s):The purpose of this study was to investigate the effects of Noggin on high glucose-induced apoptosis and insulin secretion in pancreatic beta cells. Materials and Methods: Different concentrations of glucose were used to examine their effects on INS-1 rat beta cells in vitro. When specific siRNA targeting Noggin and recombinant Noggin were added, apoptosis and insulin secretion wer...

2014 Diabetes mellitus is a chronic disease that represents a major socioeconomic burden worldwide; approximately 90–95% of patients are diagnosed as Type 2 diabetes mellitus (T2DM) among the new cases of diabetes. Pancreatic islet β-cell failure is one of the key features of T2DM, which is resultant from chronic hyperglycemia and hyperlipidemia, thus eventually leading to β-cell insulin secret...

متن کاملInsulin therapy at onset of type 2 diabetes mellitus--a new concept.
In this study, insulin therapy was initiated at onset of disease in patients whose fasting blood glucose was more than 250 mg/dl. All enrolled subjects were treated with human premixed insulin (30/70) administered subcutaneously twice daily before breakfast and before dinner. A total of 113 subjects entered the study fulfilling the inclusion criteria. Good glycaemic control was achieved in a fe...
متن کاملCorrigendum: Glucagon-like peptide-1 prevents methylglyoxal-induced apoptosis of beta cells through improving mitochondrial function and suppressing prolonged AMPK activation
Accumulation of methylglyoxal (MG) contributes to glucotoxicity and mediates beta cell apoptosis. The molecular mechanism by which GLP-1 protects MG-induced beta cell apoptosis remains unclear. Metformin is a first-line drug for treating type 2 diabetes associated with AMPK activation. However, whether metformin prevents MG-induced beta cell apoptosis is controversial. Here, we explored the sig...
